Sedentary mice treated with SLU-PP-332 showed 45–70% improvements in treadmill running endurance without any exercise training. This effect is mediated by ERRα/γ activation, which increases oxidative muscle fiber content and mitochondrial density — the same adaptations produced by endurance exercise.

Metabolic Effects

SLU-PP-332 increased mitochondrial biogenesis, enhanced fatty acid oxidation, and protected against diet-induced obesity in mice. Animals on high-fat diets gained significantly less weight when treated with SLU-PP-332, suggesting enhanced metabolic rate and fat utilization.

Muscle Fiber Type Switching

ERR activation shifted muscle fiber composition toward type I (slow-twitch, oxidative) fibers — the endurance fiber type. This mirrors the fiber type adaptation that occurs with prolonged endurance training and explains the improved exercise capacity.
