OS-01

OS-01 peptide

OS-01 is a proprietary peptide developed by OneSkin Technologies, marketed as the first topical senolytic peptide for skin. It is designed to selectively reduce senescent cells in skin tissue — aged, dysfunctional cells that accumulate with age and secrete inflammatory factors (the senescence-associated secretory phenotype, or SASP) that damage surrounding tissue. OS-01 is the active ingredient in OneSkin's topical skincare products.

Who Researches OS-01?

OS-01 is researched by people interested in evidence-based anti-aging and skin longevity through senolytic approaches. Unlike traditional anti-aging skincare (retinoids, antioxidants), OS-01 targets cellular senescence directly — the accumulation of dysfunctional cells that drives skin aging. It's relevant for researchers studying senolytics, those comparing topical versus systemic anti-aging peptides like FOXO4-DRI, and consumers looking for peptide-based skincare with peer-reviewed evidence.

What Is OS-01?

OS-01 was identified through a computational screen of peptide sequences designed to interact with senescent cell pathways. OneSkin, a San Francisco-based biotechnology company, developed the peptide as a topical senolytic — a compound that selectively eliminates senescent cells without harming healthy cells.

Senescent cells accumulate in skin with age, UV exposure, and environmental stress. While individually non-dividing, they secrete a cocktail of inflammatory cytokines, matrix metalloproteinases, and growth factors (collectively called SASP) that degrade collagen, promote inflammation, and accelerate aging of neighboring cells. By reducing senescent cell burden, OS-01 aims to interrupt this inflammatory cascade.

Mechanism of Action

Based on published research:

  • Senescent cell reduction: OS-01 decreases expression of senescence markers including p16INK4a, p21, and senescence-associated β-galactosidase (SA-β-gal) in human skin equivalents
  • SASP reduction: Lowers secretion of inflammatory factors produced by senescent cells
  • Epigenetic age reversal: OneSkin's research reported that OS-01 treatment reduced the biological (epigenetic) age of skin samples as measured by DNA methylation clocks
  • Barrier function: Improved skin barrier function and hydration in human skin models

Dosage Overview

OS-01 is available exclusively through OneSkin's commercial skincare products (face and body formulations). Applied topically twice daily to clean skin. The peptide concentration in commercial products is proprietary.

OS-01 is not available as a standalone research peptide — it is sold only as part of finished cosmetic products.

Side Effects & Safety

  • Topical application: Generally well-tolerated as a skincare product
  • No reported systemic effects: Topical application limits systemic exposure
  • Mild skin reactions: Possible irritation as with any topical product, particularly for sensitive skin
  • Long-term effects unknown: The consequences of chronic senescent cell depletion in skin over years have not been studied
