Preclinical Dosing
The original 2017 Cell paper used 5 mg/kg intraperitoneal injection in mice, administered three times per week for several weeks. No human dosing has been validated. The D-retro-inverso construction provides extended stability compared to conventional peptides.
Human-equivalent dosing has not been established through pharmacokinetic studies. Self-experimentation reports in the longevity community exist but provide no controlled data.
Preparation
FOXO4-DRI is typically supplied as lyophilized powder. Reconstitute with bacteriostatic water. The D-amino acid construction makes it more resistant to degradation than standard peptides. Use the peptide calculator for volumes.
Protocols
Mouse protocols used intermittent dosing (3× weekly). The senolytic approach generally favors intermittent "hit-and-run" dosing rather than continuous exposure — clearing senescent cells without sustained interference with normal cellular processes. No human cycling protocols exist.

- Lyophilized: -20°C for long-term
- D-amino acid peptides are more stable than L-peptides but standard cold storage is still recommended
- Reconstituted: 2–8°C
