Selective Senescent Cell Clearance
FOXO4-DRI selectively induces apoptosis in senescent cells by disrupting the FOXO4-p53 interaction that keeps these cells alive. In the original 2017 study, treatment restored fitness, fur density, and renal function in naturally aged mice — demonstrating that clearing senescent cells can reverse functional aging.
Aging Phenotype Reversal
Treated mice showed restored liver and kidney function, improved coat condition, and increased activity levels. Chemotherapy-damaged mice showed faster recovery when treated with FOXO4-DRI, suggesting accelerated clearance of therapy-induced senescent cells.
Cellular Selectivity
FOXO4-DRI's mechanism is inherently selective — it only disrupts FOXO4-p53 interactions active in senescent cells. Non-senescent cells do not rely on this interaction for survival and are therefore spared. This selectivity distinguishes FOXO4-DRI from less targeted senolytics.
