Weight Loss
Phase 3 trials in Chinese patients showed 10–15% body weight loss over 24–48 weeks. The dual GLP-1/glucagon mechanism provides weight loss through both reduced caloric intake (appetite suppression) and increased caloric expenditure (glucagon-mediated thermogenesis).
Liver Fat Reduction
The glucagon receptor component promotes hepatic fatty acid oxidation, directly addressing liver fat accumulation. This makes mazdutide potentially beneficial for MASH/NAFLD — conditions where pure GLP-1 agonists have limited liver-specific effects.
Glucose Control
Despite the glucagon component (which can raise glucose), the concurrent GLP-1 activity maintains glucose homeostasis. Phase 2 trials showed HbA1c reductions comparable to established GLP-1 agonists in type 2 diabetes patients.
